Culture of synovial cells in vitro. Notes on isolation and propagation.

چکیده
The culture of synovial cells in vitro provides a useful means for the study of connective tissue function. Vaubel (1933) first used cultures of synovium to show that mucin was secreted in the joint itself, a conclusion since confirmed by others (Kling, Levine, and Wise, 1955; Castor, 1957; Hedberg and Moritz, 1958; Fischer and Torgerson, 1960). In recent years, Castor has studied synovial culture extensively in a variety of aspects (Castor, 1959, 1960, 1962; Castor and Fries, 1961; Castor and Prince, 1963). In all these studies synovial culture has relied on the growth of cells from pieces of synovium, or "explants". In a preliminary report (Fraser and Catt, 1961), we described an alternative method for the harvest of synovial intima in a monocellular suspension suitable for culture. This paper records our further experience with the initial isolation of synovial cells, and their serial propagation beyond the stage of primary culture. Evidence is also presented to show that the new method is more likely to yield cells derived solely from the synovial intima. Certain aspects concerned with specific experimental applications will be reported in detail later.

Isolation and in vitro Characterization of Mesenchymal Stem Cells Derived from the Pulp Tissue of Human Third Molar Tooth
Background: It is still controversial that the stem cells isolated from human dental pulp meets the criteria for mesenchymal stem cells (MSCs). The aim of the present study was to examine whether or not they are MSCs, or are distinct stem cells population residing in tooth pulp. Methods: Adherent fibroblastic cells in the culture of pulp tissue from human third molars were propagated through se...
متن کاملEvaluation of the Effect of Follicle Stimulating Hormone (FSH) on Survival and Colonization of Caprine Spermatogonial Stem Cells during in Vitro Culture
Background and Objectives: Spermatogonial stem cells are specific cells that have the ability of self-renewal and differentiation. These cells play an essential role in maintaining spermatogenesis and fertility. In this regard, the present study was performed with the purpose of investigating the effect of different concentrations of follicle stimulating hormone (FSH) on in vitro colony formati...
متن کاملEffects of different culture media on optimization of primary neuronal cell culture for in vitro models assay
Background: In vitro model studies are becoming increasingly popular for experimental research designs. They include isolation and expansion of cells of a particular tissue, such as the nervous tissue which contributes to understanding the underlying mechanisms in many pathologies. It enables the scrutinization of intracellular signaling pathways responsible for cell death. OBJECTIVES: In the ...

Introduction: Monthly regeneration of endometrium after cyclical mensturation confirmed the ability of specific population of the cells that presence in the basalis layer and undergone consecutive hormonal changes that could prepared the endometrial layer for probable implantation. These cells, known as, stem cell. The aim of this study was the isolation and culture of human endometrial derive...
متن کاملAssessment of Culture Condition and In Vitro Colonization Ability of Human Spermatogonial Stem Cells: A Review Article
Spermatogenesis is a highly complex and regulated process in which germ stem cells differentiate into spermatozoa. These stem cells, called spermatogonial stem cells (SSCs), are in the base of seminiferous tubules and have the ability of self-renewal and differentiation into functional germ cells. Due to this ability, SSCs can restore spermatogenesis after testicular damage caused by cytotoxic ...
